Role of the E3 ubiquitin ligase Peli1 in regulating antitumor T cell responses

چکیده

Abstract Metabolic fitness of T cells is crucial for immune responses against infections and tumorigenesis. Both the cell receptor signal (TCR) environmental cues contribute to induction metabolic reprogramming, but underlying mechanism incompletely understood. Here, we identified E3 ubiquitin ligase Peli1 as an important regulator metabolism antitumor immunity. ablation profoundly promotes tumor rejection, associated with increased effector function number tumor-infiltrating CD4 CD8 cells. Moreover, deficiency inhibited expression PD-1 TIM-3 reduced exhaustion The Peli1-deficient display markedly stronger activities, particularly glycolysis, than wild-type controls activation mTOR signaling, stimulated by both TCR growth factors, this mediated through regulation mTORC1-inhibitory proteins, TSC1 TSC2. mediates non-degradative ubiquitination TSC1, thereby promoting stabilization TSC complex. These results establish a novel mTORC1 downstream mTORC1-mediated actions on

The Role of E3 Ubiquitin Ligase Cbl Proteins in Interleukin-2-Induced Jurkat T-Cell Activation

Interleukin- (IL-) 2 is the major growth factor for T-cell activation and proliferation. IL-2 has multiple functions in the regulation of immunological processes. Although most studies focus on T-cell immunomodulation, T-cell activation by IL-2 is the foundation of priming the feedback loop. Here, we investigated the effect of MAPK/ERK and PI3K/Akt signaling pathways on IL-2-induced cell activa...

Essential role of E3 ubiquitin ligase activity in Cbl-b-regulated T cell functions.

E3 ubiquitin ligases have been placed among the essential molecules involved in the regulation of T cell functions and T cell tolerance. However, it has never been experimentally proven in vivo whether these functions indeed depend on the catalytic E3 ligase activity. The Casitas B-cell lymphoma (Cbl) family protein Cbl-b was the first E3 ubiquitin ligase directly implicated in the activation a...

Characterization of the role of COP9 signalosome in regulating cullin E3 ubiquitin ligase activity

Cullin RING ligases (CRLs) are the largest family of cellular E3 ubiquitin ligases and mediate polyubiquitination of a number of cellular substrates. CRLs are activated via the covalent modification of the cullin protein with the ubiquitin-like protein Nedd8. This results in a conformational change in the cullin carboxy terminus that facilitates the ubiquitin transfer onto the substrate. COP9 s...

Diabetic cardiomyopathy: role of the E3 ubiquitin ligase.

Diabetic cardiomyopathy (DCM) is the leading cause of mortality in diabetes. As the number of cases of diabetes continues to rise, it is urgent to develop new strategies to protect against DCM, which is characterized by cardiac hypertrophy, increased apoptosis, fibrosis, and altered insulin metabolism. The E3 ubiquitin ligases (E3s), one component of the ubiquitin-proteasome system, play vital ...

Mechanism of Cullin3 E3 Ubiquitin Ligase Dimerization

Cullin E3 ligases are the largest family of ubiquitin ligases with diverse cellular functions. One of seven cullin proteins serves as a scaffold protein for the assembly of the multisubunit ubiquitin ligase complex. Cullin binds the RING domain protein Rbx1/Rbx2 via its C-terminus and a cullin-specific substrate adaptor protein via its N-terminus. In the Cul3 ubiquitin ligase complex, Cul3 subs...
